Transformation with Agrobacterium tumefaciens and Agrobacterium rhizogenes resulted in transgenic lines with secondary mutations and lower levels of CENH3. Transient transformation of carrot protoplasts with CRISPR/Cas9 ribonucleoprotein complexes (RNPs) resulted in transgene-free and mutated protoplasts. The transgene-free method was identified as the preferred method due to the absence of secondary mutations.</field><field name="mods.abstract">Das Zentromer-spezifische Histon 3 (CENH3) in der Karotte (Daucus carota) wurde mittels CRISPR/Cas9 mutiert. Die Transformation mit Agrobacterium tumefaciens und Agrobacterium rhizogenes führte zu transgenen Linien mit sekundären Mutationen und geringere Menge CENH3. Die transiente Transformation von Karottenprotoplasten mit CRISPR/Cas9-Ribonukleoprotein-Komplexen (RNPs) führte zu transgen-freien und mutierten Protoplasten.
