Colorectal cancer (CRC) shows heterogeneous responses to Tumor Treating Fields (TTF), a non-invasive therapy disrupting mitosis. In 21 CRC cell lines, TTF induced microtubule disruption, mitotic defects, metabolic stress, DNA damage, and apoptosis. NAALADL1 was upregulated in resistant cells; its knockdown enhanced TTF sensitivity by stabilizing microtubules and inducing G2/M arrest. Virtual screening identified Lumacaftor and Bestatin as synergistic with TTF, highlighting NAALADL1 as a promising target for personalized CRC therapy.
