Abstract: In plants the oxygenase-reaction of Rubisco produces the toxic compound 2-phosphoglycolate, which comes recycled by the photorespiratory cycle, immediately. In the plant mitochondrion, the combined action of glycine decarboxylase (GDC) and serine hydroxymethyltransferase (SHM) is essential for the photorespiratory flux. This study presents the first Arabidopsis GDC and SHM knockout mutants which distinguish from all other photorespiratory mutants by its lethality in the cotyledon stage. My results gave evidence that GDC and SHM are necessary in autotrophic as well as heterotrophic tissues and therefore have to be realized as not exclusive photorespiratory enzymes.
