In this thesis, the impact of sodium butyrate (SB) on mitochondrial and metabolic
function and lifespan was investigated. SB treatment showed a rapid and transient
increase in oxygen consumption in fly heads. Mass spectrometry analysis revealed incorporation
of labeled SB into mitochondrial pathway metabolites. An SB-independent way for mitochondrial
activation was assessed in this work. The mitochondrial membrane potential of C. elegans
was increased, using a light-activated proton pump, resulting in an extended median
lifespan, even if the intervention took part in mid- or late-staged worms.

Auswirkungen von Natriumbutyrat (SB) auf mitochondriale und metabolische Funktionen
und die Lebensspanne wurden untersucht. SB-Behandlung führte zu einem raschen und
vorübergehenden Anstieg des Sauerstoffverbrauchs in Fliegenköpfen. Massenspektrometrische
Analysen zeigten den Einbau von markiertem SB in mitochondriale Stoffwechselprodukte.
Außerdem wurde ein SB-unabhängiger Weg zur mitochondrialen Aktivierung untersucht.
Das Membranpotenzial von C. elegans wurde mittels einer lichtaktivierten Protonenpumpe
erhöht, dies verlängerte die mittlere Lebensspanne, sowohl in mittleren/späten Stadien.
