Charakterisierung der GKRP Mutationen P446L, H438Y, M344I und M344I-P446L im hepatischen Glucose- und Lipidmetabolismus

Abstract: The glucokinase regulatory protein variants H438Y, M344I, P446L and M344I-P446L were analysed. The cellular localisation of GKRP fluorescence fusion proteins in HeLa and Hepa 1-6 cells was compared to the WT and the interaction strength with GK in primary hepatocytes was investigated. Furthermore, the altered glucose uptake and triglyceride synthesis of P446L and H438Y was analysed in Hepa 1-6 cells and the influence on the cellular localisation of GK was described. Finally, the structural changes of the variants were simulated using the PyMOL software.
