In dieser Arbeit wird eine Methode zur Überwindung der Gleichgewichtslimitierung von
Transaminase katalysierten Reaktionen an zwei Beispielen betrachtet. Als Teil der
Reaktionsoptimierung werden verschiedene Amindonoren eingesetzt und die Auswirkungen
unterschiedlicher Substrat- und Donorkonzentrationen betrachtet. Nach einer erfolgreichen
Maßstabsvergrößerung liegt der Fokus in der Aufarbeitung und Isolierung des Produktes
aus einer Mischung verschiedener Amine. Aus den aufkommenden Problemen bei der Aufarbeitung
wurde das Konzept der selektiven Kristallisation entwickelt.

In this work, a concept of overcoming the limitations of transaminase catalyzed reactions
was tested for two different reactions. Part of the optimization process is the testing
of different amine donors and a variation of substrate and donor concentrations. After
a successful scale up the main focus was the downstream and isolation of the product.
As a result of these problems occurred during the downstream processing the selective
crystallization concept was developed.
