Colorectal cancer (CRC) is the third most prevalent and second deadliest cancer worldwide.
This prospective cohort study established patient-derived CRC organoids (75% success
rate) and tumor-infiltrating lymphocytes (TILs, 87.5% success rate) as preclinical
models for therapy prediction. Targeted NGS sequencing revealed TP53, APC, and KRAS
as the most frequent mutations; MSI was detected in 11% of patients. Organoid-TIL
co-culture enables exploration of the tumor microenvironment and immune evasion mechanisms,
supporting personalized immunotherapy strategies.

Die gezielte NGS-Sequenzierung identifizierte TP53, APC und KRAS als häufigste Mutationen;
Immuntherapiestrategien.
